Supported resolutions

Does HTTR support 1280 by 1024 resolution?
If so, how can I adjust this as this option is greyed out when I start the game.

You can run the game at any resolution supported by your graphics card. 1280 x 1024 is one of the default ones. If that is greyed out, then I suspect your graphics card cannot handle that. If you know it can, then try setting the resolution using the procedure on page 8 of the Reference manual. Let me know how that goes.
