I took this to mean, when it is checked off, the play is only run when it is chosen from a profile group of plays. What I am asking is, does it work? Or will checking this box simply omit the play?

Correct, the play will only be used if it is specifically listed in a group of plays.  If the situation goes to the AI, it will not be selected as a play.
 
As far as I know, it works as planned.
