I would like to replace one of my core infantry units with a Anti-tank unit. Can I do this ?

You can disband your infantry in the deployment phase, get the prestiege back and use it to buy an AT unit but if your infantry has some experiance you might not want to do that.
If you have a lot of prestiege you can just buy an AT. You can buy 'extra' core units as long as you haven't deployed up to the allowable scenario limit, you just can't might not be able to deploy all of your core units but you could hold the infantry off map and deploy the AT unit.

How do you disband a unit in the setup phase?

I think you highlight it like you are going to deploy it and hit D.
