Having an issue where one tiles is disrupting the whole supply chain, what am i missing? I have truck depots in all cities and sometimes in between.

There's not enough logistics points heading out of your capitol to that direction. Spare capacity further down the road doesn't do anything for the bottleneck, trucks can't double back to shift more cargo across a shorter distance in Shadow Empires. The SHQ's hex can easily become a bottleneck since everything sent or received has to go through there. Arguably the automatic logistics distribution here may not be optimal, it might be possible to mitigate the bottleneck by micro-optimizing the logistics, but honestly, you should just upgrade the truck depot in your capitol if you can, you're not exactly spoiled for spare logistics points overall.

EDIT: This assumes that the city on the right is the capitol (& SHQ site), which is not explicit in the image.

Upgrade the truck station on the city on the right or create a rail connection between these two cities.
