Welcome, since it looks like this is your first post on the forum. Generally, it is difficult for others to answer questions without having a CMO game file (.save file compressed using zip or something else) attached to the post. But I think new members on the forums may still have limitations on attachments to posts.

Just as a guess, you may want to check whether the Osprey actually has any cargo. Having the aircraft configured in a cargo loadout (Marines, 24x, or whatever) does not actually load any cargo; it's just an empty aircraft set up to move cargo. To transfer any cargo in a mission, there has to be cargo at the aircraft's base (see below for example) that is ordered to be loaded onto the Osprey as part of the mission. If the airbase is missing the cargo button in the unit status window on the right, it has no cargo to move. If the Osprey has no cargo button while in flight, it is flying empty.
