My question is if a unit's facing affects combat of fire resolution.

For example, if a unit receives fire from its flank or rear, will this have a different effect in casualties or cohesion, compared to receiving fire from its front?

Yes it does. A unit will receive more or less fire depending not only on its facing but also on its attitude. All extensively covered in the manual.
